Title: Expression and function of nicotinic acetylcholine receptors in the mouse brain

Nicotinic acetylcholine receptors (nAChRs) are broadly expressed throughout the brain and they are involved in the control of fundamental neural functions, including neurotransmitter release, neuronal signalling and plasticity. As nAChRs are often expressed in neurons with different and even opposing effect on neural circuits and behaviour, it is important to understand their function and regulatory mechanisms in specific neuronal types. To this aim, we create mouse models with neuronal-type-specific deletion of nAChRs to study the effects of these deletion on neuronal activity and on animal's behaviour. In addition, we plan to use CRISPR gene editing to fluorescently label endogenous nicotinic subunits and study their expression and subcellular localization in specific neurons. In our projects, we mostly use genetically modified mouse lines, preparation and stereotaxic injections of viral vectors, microscopy techniques and behavioural analysis. For specific parts of the projects, we also use neuronal cell lines for in vitro experiments.

The internship is supposed to take place during the Summer break (June to September 2024) but other time periods can be accommodated if needed.
